Glycine and Cortisol: Does the Bedtime Sleep Supplement Do Anything for Stress Hormones?

Glycine has been showing up in sleep-supplement stacks and "cortisol-lowering" product marketing for a few years now, usually as a powder or capsule taken shortly before bed. It is a real amino acid with a real, if narrow, evidence base behind its sleep effects. Whether it does anything specific to cortisol is a separate question, and one where the marketing tends to outrun the research.
Glycine is the simplest of the twenty standard amino acids and one the body already produces on its own. It functions as an inhibitory neurotransmitter in the spinal cord and brainstem, and it is also a major structural component of collagen, which is part of why it shows up in both sleep and skin-adjacent marketing. Those are legitimate, well-established roles, but they describe glycine's general physiology, not what a bedtime supplement dose specifically does to stress hormones.
The most-cited human evidence comes from a 2007 study in Sleep and Biological Rhythms, in which volunteers with unsatisfactory sleep took 3 grams of glycine before bed. Compared with placebo, glycine improved subjective sleep quality and sleep efficiency, shortened the time to fall asleep and to reach slow-wave sleep on polysomnography, and reduced next-day sleepiness, without changing overall sleep architecture.
Animal research offers a plausible mechanism for why: a 2015 study in Neuropsychopharmacology found that in mice, glycine promoted sleep and lowered core body temperature through NMDA receptors in the suprachiasmatic nucleus, the brain's central circadian clock, likely by encouraging heat loss through the skin's blood vessels. That is a mechanistic, animal-level finding, not direct proof of the same pathway driving results in humans, but it fits with human research showing glycine users reach their core-temperature dip, and therefore sleep onset, somewhat faster.
On cortisol specifically, the evidence is thin. No well-controlled human trial has directly demonstrated that glycine supplementation lowers cortisol output or reshapes its daily rhythm. The more defensible version of the claim is indirect: poor sleep is independently associated with flatter, less healthy cortisol rhythms in research on sleep and HPA axis function, so anything that genuinely improves sleep quality could plausibly support better cortisol regulation over time. That is a reasonable hypothesis, not something glycine has been shown to do directly, and the distinction matters when a product label states it as settled fact.
For someone curious about trying it, 3 grams before bed is the dose actually studied in the human trial, and glycine is generally considered low-risk, though anyone on medication or with a health condition should check with a doctor first. The realistic expectation is a modest sleep-quality aid with early supporting research behind it, not a direct cortisol treatment and not a proven skin fix, whatever a supplement bottle's claims might suggest.
